Application Notes

Immunohistochemistry with formalin fixed paraffin embedded tissues: 4-11 μg/mL Antigen retrieval: Steam slides in 0.01 M sodium citrate buffer, pH 6.0, at 99-100 °C for 20 min. Remove from heat and let stand at room temperature in buffer for 20 min. Rinse in 1x TBS with Tween (TBST) for 1 min. at room temperature.
Other applications not tested.

Background
- GPR65 (TDAG8), a Lysophospholipid/Lysosphingolipid Receptor, is a molecular target for psychosine, a toxic lipid formed by the breakdown of galactosylceramide (cerebroside). A build-up of psychosine, caused by the lack of galactosylceramidase, results in the death of the myelin-synthesizing cells, oligodendrocytes, which is associated with Krabbe disease. Recent evidence suggests that GPR65 is one of the proton-sensing GPCRs coupling to adenylyl cyclase and psychosine and its related lysosphingolipids behave as if they were antagonists against protein-sensing receptors, including TDAG8, GPR4 and OGR1 GPR65 has been reported in human in peripheral blood leukocytes, spleen, lymph node, and thymus. ESTs have been isolated from human B-cell/lung/testis, brain/lung/testis, pancreas/spleen, lung/spleen, leukocyte, embryo, kidney, liver/spleen, thyroid, and vessel libraries.Synonyms: G-protein coupled receptor 65, T-cell death-associated protein 8, TDAG8
