    Background

    Lysosomes are membrane-bound vacuoles which play a critical role in endocytosis, synthesis and assembly of acid hydrolases. These sites are also site for digestion of foreign materials and for specialized autolytic cellular processes. Lysosome associated membrane proteins 1 and 2 (LAMP1/CD107a and LAMP2/CD107b) are members of the lysosome associated membrane glycoprotein family and are well known lysosomal markers. The disulfide arrangement is common to both members of this family of lysosomal membrane glycoproteins as observed by conservation of cysteine residues among lysosome-associated membrane proteins 1 and 2. LAMP1, also known as CD107a, is a heavily glycosylated membrane protein which has an important role in normal lysosomal function. This glycoprotein provides selectins with carbohydrate ligands and is a crucial mediator for cellular adhesion by interacting with carbohydrate receptors.It contains a putative signal peptide, 18 sites for N-linked glycosylation, a single membrane-spanning segment and a short (11 amino acid) cytosolic tail. LAMP1 plays a role in tumor cell invasion and metastasis. It is involved in lysosomal membrane protection and in lectin-mediated cell adhesion, and is also a marker of degranulation on lymphocytes such as CD8+ and NK cells. LAMP1, also called LAMPA, LGP120 or CD107a, is a member of a family of membrane glycoproteins and plays an important role in tumor cell metastasis. LAMP1 expression correlates with the metastatic potential of some tumor cells, and with neurons and glial cells surrounding plaques in Alzheimer's patients. LAMP1 also serve as a marker of mast cell activation and regulate the biogenesis of pigment melanin. LAMP1 is a 100-120 kDa type I integral lysosomal membrane glycoprotein that shuffles between lysosomes, phagosomes, and the plasma membranes. It is a transmembrane protein which is heavily glycosylated at the amino-terminal luminal side of the lysosomal inner leaflet, protecting the proteins from proteolysis. The carboxy terminus of LAMP1 is exposed to the cytoplasm and contains a tyrosine sorting motif that targets LAMP to lysosomal membranes. LAMP1 and LAMP2 are 37 % homologous in their protein sequences and both are involved in regulating lysosomal motility during lysosome-phagosome fusion and cholesterol trafficking. LAMP-1 is expressed on the surface of thrombin-activated but not resting platelets, and is involved in the adhesive, prothrombic properties of these cells. It is also involved in maintaining lysosome acidity and protecting the lysosomal membranes from auto digestion, and their expression is increased in patients with lysosomal storage disorders.The gene for LAMP1 is present on chromosome 13q34
