Western Blotting (WB), ELISA, Immunohistochemistry (IHC), Immunoprecipitation (IP)
    Cross-Reactivity
    Mouse (Murine), Rat (Rattus), Monkey
    Cross-Reactivity (Details)
The antibody does not cross react to any other cellular protein.
    Characteristics
ROR1-selective antibodies were generated against a peptide taken the C-terminal region of the ROR1 protein. The ROR1-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The anti-ROR1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. antibodies-online.com provides ROR1 Western blot positive control samples in SDS-PAGE sample buffer.

  • Application Notes
Antibodies were tested in ELISA and western blotting applications at 1:500 dilution using ABIN1686551 samples. Antibody dilutions for these antibodies are for reference only, investigators are expected to determine the optimal conditions. Application of this antibody in other protocols has not yet tested.
    WB: > 1:500
    IMM & IP pull-down assays: n.d.
    IHC: n.d.
Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. n.d. not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application.
The antibody labels a strong band of ROR1 at 113 kDa in ABIN1686551 samples and in other cancer cell lines.

    Background
The Receptor tyrosine kinase-like Orphan Receptor (ROR) proteins are conserved tyrosine kinase receptors that pattern tissues and organs during embryonic development and in adulthood. ROR family proteins are ligand-dependent transcription factors that are required for skeleton and neuronal development, and modulate cell migration, cell polarity, and convergent extension. RORs act as Wnt receptors signaling via novel non-canonical Wnt pathways mediated by the adapter protein disheveled and the non-receptor tyrosine kinase Src. RORs can either activate or repress Wnt target expression depending on the cellular context and can also modulate signal transduction by sequestering Wnt ligands away from their signaling receptors. The mammalian ROR family of receptor tyrosine kinases consists of two structurally related proteins, ROR1 and ROR2. ROR1 is an orphan cell surface receptor with strong homology to the tyrosine kinase domain of growth factor receptors, in particular the Trk family. Increased expression of this gene is associated with B-cell chronic lymphocytic leukemia and alternative splicing results in multiple transcript variants encoding different isoforms. ROR1 is expressed by many tissues during embryogenesis, some B-cell malignancies, and various cancer cell lines with high levels of phosphorylated AKT (p-AKT) and phosphorylated cAMP response element binding-factor (p-CREB). ROR1 plays a functional role in promoting tumor cell growth, suggesting that this orphan-receptor tyrosine-kinase-like protein may be a potential target for therapy directed against a variety of human cancers. Silencing the expression of ROR1 in human breast cancer cell lines impaired their growth in vitro and also in immune-deficient mice. High-level expression of ROR1 is found in breast adenocarcinoma associated with aggressive disease indicating that it may be a potential target for breast cancer therapy. The ROR1 gene is mapped to chromosome 1p12-p32
