    ELISA. Western blot (1/500-1/1,000). Immunohistochemistry on Paraffin Sections (2.5 μg/mL). This APP antibody was validated for use in immunohistochemistry on a panel of 21formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigenretrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides wereincubated with biotinylated secondary antibody, followed by alkalinephosphatase-streptavidin and chromogen.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Amyloid precursor protein (APP) is the precursor molecule whose proteolysis generates amyloid beta (Aβ), a 39- to 42- amino acid peptide and this amyloid fibrillar form is the primary component of amyloid plaques found in the brains of Alzheimer's diseases patients. APP is an integral membrane protein that is phosphorylated in the cytoplasmic and extracellular domains. It has been reported that cell-surface APP plays a role in neurite extension of primary cultured hippocampal neurons. The large extracellular domain of APP is also reported to bind extracellular matrix molecules such as heparin, laminin, and collagen, which can mediate cell adhesion and neurite outgrowth.
