    Extracellular Domain

    Purpose

    Rabbit antibody to TrkA

    Specificity

    Specific for TrkA.

    Cross-Reactivity

    Mouse, Rat

    Cross-Reactivity (Details)

    Other species not yet tested.

    Purification

    IgG

    Immunogen

A synthetic peptide from the extracellular domain of rat TrkA conjugated to blue carrier protein was used as the antigen. The peptide is homologous in mouse.

    Isotype

    IgG
  • Application Notes

IHC WB. A concentration of 10-50 μg,ml is recommended. The optimal concentration should be determined by the end user.

    Restrictions

    For Research Use only
  • Format

    Lyophilized

    Reconstitution

Reconstitute in 500 µL of sterile water. Centrifuge to remove any insoluble material.

    Handling Advice

    Avoid freeze and thaw cycles.

    Storage

    4 °C,-20 °C

    Storage Comment

Maintain the lyophilised/reconstituted antibodies frozen at -20C for long term storage and refrigerated at 2-8C for a shorter term. When reconstituting glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

    Alternative Name

    TrkA

    Background

FUNCTION: Required for high-affinity binding to nerve growth factor (NGF) neurotrophin-3 and neurotrophin-4/5 but not brain-derived neurotrophic factor (BDNF). Known substrates for the Trk receptors are SHC1 PI 3-kinase and PLC-gamma-1. Has a crucial role in the development and function of the nociceptive reception system as well as establishment of thermal regulation via sweating. Activates ERK1 by either SHC1- or PLC-gamma-1-dependent signaling pathway. CATALYTIC ACTIVITY: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate. SUBUNIT: Exists in a dynamic equilibrium between monomeric (low affinity) and dimeric (high affinity) structures. Binds SH2B2. Interacts with SQSTM1 which bridges NTRK1 to NGFR. Interacts with NGFR and ARMS. Can form a ternary complex with NGFR and ARMS and this complex is affected by the expression levels of ARMS. An increase in ARMS expression leads to a decreased association of NGFR and NTRK1. SUBCELLULAR LOCATION: Cell membrane, Single-pass type I membrane protein. Note: Endocytosed to the endosomes upon treatment of cells with NGF. TISSUE SPECIFICITY: Isoform Trka-II is primarily expressed in neuronal cells, isoform Trka-I is found in non-neuronal tissues.
