Purpose
- MLKL is a protein encoded by the MLKL gene which is approximately 54,4 kDa. MLKL is localised to the cell membrane and cytoplasm. It is involved in the dimerization of procaspase-8, the TNF signalling pathway, apoptosis and autophagy. This protein falls under the protein kinase superfamily. It plays a critical role in tumour necrosis factor-induced necroptosis which is a programmed cell death process. It does this via interaction with the receptor-interacting protein 3 which is a key signalling molecule in necroptosis pathway. MLKL is expressed in the heart, intestine and lung. STJ97776 was developed from clone 6F8 and was affinity-purified from mouse ascites by affinity-chromatography using specific immunogen MLKL. This primary antibody specifically binds to endogenous MLKL protein which only binds about S358 when S358 is phosphorylated.
