Description: Collapsin response mediator protein 2 (CRMP-2, DRP-2), is a kinesin-binding protein and a member of the CRMP family, a novel family of microtubule-associated proteins (MAPs). This family consists of 5 highly conserved phosphorylated proteins: CRMP1-CRMP5. CRMP-2 was originally identified in the regulation of microtubule dimerization. CRMP-2 is the first member of the CRMP family identified in primary neuronal cells1,2.Physiological functions of CRMP-2 protein include regulation of cell surface receptor endocytosis, kinesin-dependent axonal transport, growth cone collapse, polarity and differentiation of developing neurons and neurite outgrowth and the regulation of microtubule dynamics2.Studies suggest that CRMP-2 can modulate neuronal viability through the PI3K/mTOR/S6K pathway. In addition, CRMP-2 expression has been detected in non-neuronal cells such as leukocytes, fibroblasts and neuroblastoma1.Phosphorylation of CRMP-2 on specific Thr and Ser residues inactivates its activity, leading to inhibition of axonal growth and neuronal polarity. Knockdown of CRMP-2 shortens the total dendritic length and decreases dendritic branching3.
