    Specificity

    This antibody binds the MUC1, a tumor associated antigen expressed on many breast cancer cells. This antibody does not show reactivity towards fibroblasts, lymphoblastoid cells, and a large number of epithelial cell lines of non-breast origin. This antibody recognizes the 'PDTR' epitope of mucin.

  • Application Notes

    This antibody reacts with most of the breast cancer cells lines and with epithelial cells of human milk. The antibody also reacts with neoplasms of epithelial origin, particularly carcinomas of breast, ovary, cervix, bronchus and gastrointestinal tract. This antibody was used for the study of expression of epithelial antigen in normal and disordered cervical squamous epithelium using immunohistological methods (PMID: 6315048). This antibody can recognize the epithelial membrane antigen in a radioimmunoassay. Immunofluorescent staining of cultures of human breast cells with these antibodies demonstrated that they are heterogeneous in their distribution (PMID: 6086018). HMFG1 and HMFG2 recognize different epitopes of the millk fat globule (PMID: 6086018, 2424847). This antibody was used for the study of tumor associated antigenic epitopes purified from human skimmed milk using immunoaffinity chromatography, immunoprecipitation and immunoblot techniques (PMID: 2443241). The reshaped humanized antibody, HuHMFGl, showed no difference in relative affinity for its antigen compared with the parent mouse HMFG1 (PMID: 7682986). This mAb reacts with P1-24 synthetic peptide (PDTRPAPGSTAPPAHGVTSAPDTR) representing the repeating amino acid sequence of the mucin core protein and also reacted strongly with ovarian carcinoma-associated sebaceous gland antigen (SGA) by direct binding and in a sandwich ELISA assay with OM-1 as the capture Mab (PMID: 1697426).
