  • Purpose

    Anti-XCR1 [MARX10], Rabbit IgG, kappa

    Specificity

    This antibody binds mouse XCR1, which is a receptor for chemokines SCYC1 and SCYC2 and expressed on 80 % of CD8+ the dendritic cells. It transduces a signal by increasing the intracellular calcium ions level. This antibody is also reported to cross-reacts with rat CXR1.

    Application Notes original Clone: This antibody was used to stain cryostat sections (12 μm) of spleens from C57BL/6 WT and B6.XCR1-lacZ mice. It was found that this antibody could stain 20 % of splenic conventional dendritic cells in C57BL/6 WT mice but did not give a signal in homozygous B6.XCR1-lacZ animals, which lack the XCR1 receptor. This antibody was also used to subsets of dendritic cells that express XCR1 using flow cytometry (Bachem et al., 2012, PMID: 22826713). Digoxigenin (Dig) coupled MARX10 was injected into C57BL/6 mice for in vivo labeling of XCR1+ dendritic cells. Recombinant ovalbumin (OVA) coupled MARX10 (MARX10-OVA) was used to measure antibody levels in C57BL/6 and homozygous B6.XCR1-lacZ mice serum following injection and for an in vivo cytotoxicity assay, MARX10-OVA was found to be highly effective in inducing CD8+ T cell cytotoxicity in vivo (Hartung et al., 2015, PMID: 25520399). This antibody has been added to the Antibody Registry (RRID: AB_3668949).
