    Purpose

    Anti-Glypican 3 [7H1], Rabbit IgG, kappa

    Specificity

    This antibody binds an epitope between amino acids 25 to 137 of the human glypican 3 (GPC3). This antibody cross reacts with cynomolgus monkey, rat and mouse GPC3. Glypican-3 is an oncofetal antigen that belongs to the glypican family of glycosyl-phosphatidylinositol-anchored heparin sulfate proteoglycans. It negatively regulates the hedgehog signaling pathway and positively regulates the canonical Wnt signaling pathway. Multiple studies have revealed that GPC-3 is overexpressed in hepatocellular carcinoma (HCC).

  • Application Notes

    This antibody can bind human glypican 3 expressed on the surface of 293S cells, HepG2 X1 cells and JHH7 cells in a flow cytometric assay. The binding characterization of this antibody to huGPC3, cynoGPC3, and HepG2 cells was done using ELISA and immunohistochemistry. This antibody strongly reacts with hepatic cancer tissue microarray, JHH cells, HepG2 cells, and cells stably transfected with GPC3 in immunohistochemistry. This antibody also detects human, cynomolgus monkey, rat, and mouse GPC3 by western blot. This antibody was also used in the generation of an antibody drug conjugate with maleimide acetal PNU-159682 or monomethyl disulfide N10-linked PBD. The ADCC activity of the antibody drug conjugate was investigated using a human HepG2 X1 and JHH7 xenograft model and substantial tumor growth inhibition was achieved with 7H1-disulfide-PBD (US9926377).

    This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format for improved compatibility with existing reagents assays and techniques.
