Background
-
Background_Plain: DPPA2 and DPPA4 (developmental pluripotency associated 2 and 4) are small heterodimerizing nuclear proteins linked in the biological processes of transcription and transcription regulation. The encoded proteins have a scaffold-attachment factor A/B, acinus and PIAS (SAP) domain that binds DNA and is thought to modify chromatin. The nuclear factor encoded by the gene is involved in the maintenance of pluripotency in stem cells and is found to be essential for embryogenesis. DPPA2 and DPPA4 are required to maintain H3K4me3 and H3K27me3 at important bivalent promoters depicted in pluripotent cells by low H3K4me3 levels and initiating but not elongating RNA polymerase II. This gene is thought to play an important role in tumor progression because when mutated it is highly expressed in embryonal carcinomas, pluripotent germ cell tumors, and other cancers.
