    Protocol

    The microtiter plate provided in this kit has been pre-coated with an antibody specific to IL-13. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for IL-13 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain IL-13, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of IL-13 in the samples is then determined by comparing the O.D. of the samples to the standard curve. 2

    Background

    Interleukin 13 (IL-13) is produced primarily by activated T lymphocytes. It shows approximately 30% amino acid (aa) sequence identity with IL-4 and shares many biological properties with IL-4. IL-13 has multiple effects on the differentiation and functions of monocytes/macrophages. It can suppress the cytotoxic functions of monocytes/macrophages and the production of proinflammatory cytokines by monocytes/macrophages. Although rat or mouse IL-13 has effects similar to those of IL-4 on rat B cells, mouse IL-13 has no effect on mouse B cells. Unlike IL-4, which is a growth and differentiation factor for rat and mouse T cells, IL-13 has no effects on either rat or mouse T cells. The biological effects of IL-13 are mediated by specific high-affinity cell surface receptor complexes. The functional IL-13 receptor complex has been shown to consist of the low-affinity IL-13 receptor α chain and the IL-4 receptor α subunit (IL-4 Rα). This IL-13 receptor complex may also serve as an alternate high-affinity IL-4 receptor complex in IL-4 responsive cells that lack the γ chain.
