  • Application Notes

HTRF assay for KIT (546-976) activity 1 uM TK substrate was incubated with different concentrations of KIT (546-976) protein in a 10 µL reaction system containing 1xEnzymatic Buffer, 5 mM MgCl2, 1 mM DTT, 1 mM MnCl2, 5 nM SEB and 100 uM ATP for 1 hour. Then 10 µL detection reagents containing TK antibody (1:2) and SA-XL665 (1:100) diluted with 1x Detection Buffer were added and incubated with the reactions for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.

    Handling Advice

Avoid repeated freeze/thaw cycles, keep on ice when not in storage

    Storage

    -80 °C

    Storage Comment

Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.

    Background

Protein Background: MST2 also known as STK3 is a stress-activated, pro-apoptotic kinase which, following caspase cleavage, enters the nucleus and induces chromatin condensation followed by inter nucleosomal DNA fragmentation. Key component of the Hippo signaling pathway which plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein STK3/MST2 and STK4/MST1, in complex with its regulatory protein SAV1, MST2 phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/ TAZ. Phosphorylation of YAP1 by LATS2 inhibits its translocation into the nucleus to regulate cellular genes important for cell proliferation, cell death, and cell migration. STK3/MST2 and STK4/MST1 are required to repress proliferation of mature hepatocytes, to prevent activation of facultative adult liver stem cells (oval cells), and to inhibit tumor formation. MST2 phosphorylates NEK2 and plays a role in centrosome disjunction by regulating the localization of NEK2 to centrosome, and its ability to phosphorylate CROCC and CEP250.

Short Description: Recombinant MST2 protein was expressed in baculovirus expression system as the full-length protein (accession number NP_006272.2) with a N-terminal FLAG tag. The molecular weight of the protein is 57.6 kDa.
