Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhFGF-2 (0.3 ng/mL), a concentration of 0.25-0.40 μg/mL of this antibody is required. ELISA: (Indirect): To detect hFGF-2 by indirect ELISA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant hFGF-2. (Sandwich): To detect hFGF-2 by sandwich ELISA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with Biotinylated Anti-Human FGF-2 (PP1007B1, PP1007B2) as adetection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant hFGF-2. Western Blot: To detect hFGF-2 by Western Blot analysis this antibody can be used at aconcentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hFGF-2 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

FGF basic (FGF2) is a member of the fibroblast growth factor (FGF) family. FGF family members bind heparin and possess broad mitogenic and angiogenic activities. FGF2 is involved in diverse biological processes, such as limb and nervous system development, wound healing, and tumor growth. FGF2 mRNA contains multiple polyadenylation sites, and is alternatively translated from AUG and non-AUG (CUG) initiation codons resulting in five different isoforms with distinct properties.
