Specificity The monoclonal antibody 6D1 reacts with rat CD59, a 20 kDa glycoprotein also known as protectin. CD59 is expressed on a variety of cell types, including neutrophils, monocytes, endothelial cells, and erythrocytes. This GPI-anchored glycoprotein inhibits complement-mediated cell lysis by binding to C8 and C9, which blocks incorporation of C9 Molecules into the membrane attack complex required for osmolytic pore formation. Studies have shown that CD59 is also involved in signal transduction leading to T cell activation.

The 6D1 antibody has been reported to have antagonist activity.

Application Notes Applications reported: This 6D1 antibody has been reported for use in flow cytometric analysis.

Applications tested: This 6D1 antibody has been tested by flow cytometric anaysis of rat bone marrow cells. This can be used at less than or equal to 1 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10 5 to 10 8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. PerCP-eFluor® 710 can be used in place of PE-Cy5, PE-Cy5.5 or PerCP-Cy5.5. PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Please make sure that your instrument is capable of detecting this fluorochrome. For a filter configuration, we recommend using the 685 LP dichroic mirror and 710/40 band pass filter, however the 695/40 band pass filter is an acceptable alternative. Fixation: Samples can be stored in IC Fixation Buffer (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution for up to 3 days in the dark at 4 °C with minimal impact on brightness and FRET efficiency/compensation.
