Nerve growth factor (NGF) stimulates division and differentiation of sympathetic andembryonic sensory neurons. NGF is thought to have a profound effect on thedevelopment and maintenance of sympathetic and embryonic sensory neurons (1). NGFactivity isolated from the male mouse submaxillary gland (MSG) consists of three typesof subunits, alpha, beta and gamma, which specifically interact to form a 7S,approximately 130,000-molecular weight (Mr) complex. The 7S complex contains twoidentical 118-amino acid beta-chains, which are solely responsible for the nervegrowth-stimulating activity of NGF. While NGF is found in almost all vertebrates, mostresearch has focused on murine NGF, as the mouse male submaxillary gland containshigher levels of this polypeptide than other tissues (2). . Washing buffer:TBST washing buffer: 1XTBS/0.1 % Tween-20To prepare stock solution of 10X TBS: add 24.2 g Trizma base and 80 g sodium chloride to 1L of dH2O. Adjust pH to 7.6.Working solution. 1XTBST/0.1 % Tween-20: add 100ml 10XTBS to 900 mL dH2O. Add 1 mL Tween-20 and mix well.1.
