Application Notes
-
Direct ELISA: To detect hBD-2 by direct ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant hBD-2. Sandwich ELISA: To detect hBD-2 by Sandwich ELISA (using 100 μL/well antibody solution)a concentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with a compatible polyclonal Anti-Human BD-2 (PP1125P1 orPP1125P2) as a Capture antibody, allows the detection of at least 0.2-0.4 ng/well ofrecombinant hBD-2. Western Blot: To detect hBD-2 by Western Blot analysis this antibody can be used at aconcentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hBD-2 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user. -

Background
- This antibiotic peptide is locally regulated by inflammation. Defensins form a family of microbicidal and cytotoxic peptides made by neutrophils. Members of the defensin family are highly similar in protein sequence. hBD-2 is a antimicrobial peptide, which belongs to the distinct family of beta-defensins.Synonyms: BD-2, Beta-defensin 2, Beta-defensin 4A, DEFB102, DEFB2, DEFB4A, DEFB4B, Skin-antimicrobial peptide 1, hBD-2
