Application Notes

Suitable for ELISA, Immunoprecipitation, Immunoblotting and other immunologicalmethods requiring high titer and specificity. Recommended Dilutions: This product was assayed by immunoblot and found to bereactive against p27 from cell lysates and E. coli produced fusion proteins at a dilution of1: 2000 followed by reaction with Peroxidase conjugated Affinity Purified anti-Rabbit IgG[H&L] (Goat). Expect a band at approx. 27 kDa in size corresponding to p27 by western blotting inthe appropriate cell lysate or extract. Anti-p27 is suitable for the detection by immunoblot of human, rat and mouse p27. This antisera is suitable for Immmunoprecipitation of in vitro translated protein and celllysates (C212, 3T3-L1) and will co-precipitate associated proteins.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user. -

Background
- P27 Kip is a cyclin-dependent kinase inhibitor that shares a limited similarity with CDK inhibitor CDKN1A/p21. p27 binds to and prevents the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes, and thus controls the cell cycle progression at G1. The degradation of this protein, which is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state.Synonyms: Cyclin-dependent kinase inhibitor 1B, Cyclin-dependent kinase inhibitor p27, p27Kip1
