    Specificity

    The antibody detects endogenous level of p95/NBS1 only when phosphorylated at serine 343.

    Purification

    The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatogramphy using non-phosphopeptide corresponding to the phosphorylation site.

    Immunogen

    Peptide sequence around phosphorylation site of pSer343 (S-L-S (p) -Q-G) derived from Human p95, NBS1. Antibodies were produced by immunizing rabbits with synthetic phosphopeptide and KLH conjugates.

    Background

    Mutations in p95/NBS1 gene are associated with Nijmegen breakage syndrome, an autosomal recessive chromosomal instability syndrome characterized by microcephaly, growth retardation, immunodeficiency, and cancer predisposition. The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. This gene product is thought to be involved in DNA double-strand break repair and DNA damage-induced checkpoint activation.
