    Protocol

    human PAI-1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PAI-1 has been precoated onto 96-well plates. Standards(sf21, S22-P402) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PAI-1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PAI-1 amount of sample captured in plate.

    Assay Procedure

    Aliquot 0.1 mL per well of the 10000pg/mL, 5000pg/mL, 2500pg/mL, 1250pg/mL, 625pg/mL, 312pg/mL, 156pg/mL human PAI-1 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, cell lysates, serum or plasma(heparin, EDTA, citrate ) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each human PAI-1 standard solution and each sample be measured in duplicate.

    Background

    Protein Function: Serine protease inhibitor. This inhibitor acts as 'bait' for tissue plasminogen activator, urokinase, protein C and matriptase-3/TMPRSS7. Its rapid interaction with PLAT may function as a major control point in the regulation of fibrinolysis. .

    Background: Plasminogen activator inhibitor-1 is the principal inhibitor of tissue plasminogen activator(tPA) and urokinase(uPA), the activators of plasminogen and hence fibrinolysis(the physiological breakdown of blood clots). It is a serine protease inhibitor(serpin) protein(SERPINE1). PAI-1 is mainly produced by the endothelium(cells lining blood vessels), but is also secreted by other tissue types, such as adipose tissue. The PAI-1 gene is located on the seventh chromosome(7q21.3-q22).1 Congenital deficiency of PAI-1 leads to a hemorrhagic diathesis(a tendency to hemorrhage). PAI-1 is present in increased levels in various disease states(such as a number of forms of cancer), as well as in obesity and the metabolic syndrome. It has been linked to the increased occurrence of thrombosis in patients with these conditions. Additionally, it has been shown to interact with ORM1.2 The standard product used in this kit is recombinant human PAI-1 with the molecular mass of 43KDa.
