BoxAB from HMGB1 includes both BoxA and BoxB domains connected as in the natural sequence but lacks the negatively charged C-terminal tail. This tailless form retains the structural integrity and receptor-binding capacity of full-length HMGB1, while showing altered functional behavior in redox-dependent signaling and immune activation.
BoxAB is ideal for studying the mechanisms of HMGB1-mediated inflammation, receptor binding (RAGE, TLR2, TLR4), and DNA interaction without interference from the acidic tail. The protein is fully compatible with in vitro biochemical assays, cell culture studies, and receptor-ligand binding analyses.
The product is produced in E.coli from an expression plasmid coding for the human HMGB1 sequence (amino acids 1–176) without any tags or modifications, ensuring natural folding and charge distribution. -
Purification
- Purified. Oxidation of cysteine 106 makes the protein inactive. To avoid cysteine oxidation DTT 0.5 mM is added during protein purification.
