  • Application Notes
    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhFlt-3 Ligand (15 ng/mL), a concentration of 0.25-0.75 μg/mL of this antibody is required. ELISA: (Indirect): To detect hFlt-3 Ligand by indirect ELISA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant hFlt-3 Ligand. (Sandwich): To detect hFlt-3 Ligand by sandwich ELISA (using 100 μL/well antibodysolution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with Biotinylated Anti-Human Flt-3 Ligand (PP1008B1,PP1008B2) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well ofrecombinant hFlt-3 Ligand. Western Blot: To detect hFlt-3 Ligand by Western Blot analysis this antibody can be used ata concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hFlt-3 Ligand is 1.5-3.0 ng/lane, under either reducingor non-reducing conditions.

    Background
    Flt3 ligand is a ligand for the FLT3 tyrosine kinase receptor and belongs to a small group of growth factors that regulate proliferation of early hematopoietic cells. Multiple isoforms of Flt3 ligand have been identified. The predominant form is the transmembrane form, which is biologically active on the cell surface. When proteolytically cleaved the transmembrane isoform generates a soluble form, which is also biologically active. Flt3 ligand binds to cells expressing the tyrosine kinase receptor Flt3. Flt3 ligand alone cannot stimulate proliferation, but synergizes well with other CSFs and interleukins to induce growth and differentiation.Synonyms: FLT3LG, Flt3L, SL cytokine, fms-related tyrosine kinase 3 ligand
