Application Notes
Immunohistochemistry on paraffin sections: 1: 100 - 1: 300, requires protein digestionpretreatment of paraffin sections e. g. trypsin or pronase. ELISA: 1: 10000 - 1: 50000. Western Blot: 1: 1000 - 1: 10000, detects a band of approximately 52 kDa. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Handling Advice
Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Pdcd4 interacts with the RNA helicase eIF4A and the scaffolding protein eIF4G, two components of the translation initiation complex, and thus inhibits protein synthesis. Pdcd4 has also been shown to inhibit transcription factor activator protein-1 (AP-1), which regulates several events required for cell invasion, including expression of matrix metalloproteinases and cell motility. Pdcd4 localizes to the nucleus in proliferating cells, and its expression is modulated by cytokines in natural killer and T cells.
