Specificity:

This antibody is reactive with rat CD4 domain 3 & 4, and is partly competitive with OX-67 & 69.

Application Notes original Clone: This antibody binds to rat CD4. Upon challenge with HIV-1, this antibody was able to significantly inhibit viral entry into HeLa cells expressing a mutant of rat CD4 that binds gp120 (SImon, 1997). Similarly, pre-binding of gp120 resulted in a 50 % reduction in binding of this antibody, indicating epitope masking. Analogous effects on viral entry and synctium formation are observed with MAbs against human CD4.

Characteristics: This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2a format, for improved compatibility with existing reagents, assays and techniques.

Immunogen: This antibody was prepared by subcutaneously injecting BALB/c mice with a soluble form of rat CD4, consisting of domains 3 and 4. A boost of a mixture of free sCD4 and sCD4 bound to cells was administered intravenously.
