Background | The epidermal growth factor (EGFR, phospho EGFR), also known as ErbB1 and HER1, is a receptor tyrosine kinase (RTK) and a known oncogenic driver of several cancer types including lung cancer (1), breast cancer (2), and glioblastoma (3) and inhibitors of EGFR have been the most successful examples of targeted therapies including antibody therapeutics (e.g. trastuzumab and cetuximab) as well as small-molecule inhibitors erlotinib and lapatinib (4). Ligand binding induces multiple EGFR auto-phosphorylation including Tyr1068. This phosphorylation recruits growth factor receptor-bound protein 2 (Grb2) which mediates downstream modulation of EGFR signaling including activation of signaling cascades through interaction of Grb2 SH3 domains with the Ras guanine exchange factor, son of sevenless homolog (SOS) or downregulation of phospho EGFR signaling through internalization and ubiquitination-targeted degradation (5). |
