    Specific Activity: 1.3 - 6.7 x 10^7 Units /mg
    ED50: 150-750 pg/ml
    Observed dose response: 64 fold.
A Unit is defined as the amount of material required to stimulate a half-maximal response at cytokine saturation
    Characteristics
Recombinant mouse IL-6 protein is supplied as a frozen liquid comprised of 0.22 µm sterile-filtered Dulbecco’s PBS (pH 7.2) containing 1.0 mg/ml biotechnology grade, low endotoxin bovine serum albumin, and glycerol, with no preservatives. The recombinant mouse IL-6 was purified by immunoaffinity chromatography and was found to be > 95% pure by SDS-PAGE analysis, and an absorbance assay based on the Beers-Lambert law.

  • Application Notes
ELISA Standard: The recombinant mouse IL-6 is useful as a quantitative standard for measuring mouse IL-6 protein levels in an IL-6 specific sandwich ELISA with the purified MP5-20F3 antibody (Cat. No. 554400) as a capture antibody and the biotinylated MP5-32C11 (Cat. No. 554402) as the detection antibody. To obtain linear standard curves, doubling dilutions of this mouse IL-6 standard from ~2,000 to 15 pg/ml should be included in each ELISA plate.
Ligand Blocking Control for Immunofluorescent Staining and Flow Cytometric Analysis: The recombinant mouse IL-6 can be used as a blocking control to demonstrate specificity of IL-6 staining (for instance staining with the PE-MP5-20F3 antibody, Cat. No. 554401). The intracellular cytokine staining technique and use of blocking controls are described in detail by C. Prussin and D. Metcalfe.

    Handling Advice
Interleukin-6 (IL-6), also known as IFN-β2, BSF-2, and HPGF, is a multifunctional cytokine which regulates immune responses, acute-phase reactions and hematopoiesis. Reported cellular targets for IL-6 action include hepatocytes, B and T lymphocytes, neurons, tumor cells and multipotent hematopoietic cells. Mouse IL-6 is a 21.7 kD protein containing 187 amino acid residues.

    Storage Comment
Store product at -80°C prior to use or for long term storage of stock solutions. Rapidly thaw and quick-spin product prior to use. Avoid multiple freeze-thaws of product. This preparation contains no preservatives, thus it should be handled under aseptic conditions.
