  • Application Notes
    Neutralisation: To yield one-half maximal inhibition [ND50] of the biological activity ofhPDGF-AA (3.0 ng/mL), a concentration of 0.03 - 0.05 μg/mL of this antibody is required. Immunohistochemistry: A starting dilution of 1 μg/mL is recommended for frozen sections. ELISA: To detect hPDGF-AA by direct ELISA (using 100l/well antibody solution) aconcentration of at least 0.5 g/ ml of this antibody is required. This antigen affinitypurified antibody, in conjunction with compatible secondary reagents, allows the detectionof 0.2 - 0.4 ng/well of recombinant hPDGF-AA. Western Blot: To detect hPDGF-AA by Western Blot analysis this antibody can be used at aconcentration of 0.1 - 0.5 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hPDGF-AA is 1.5 -3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.

    Background
    PDGF is a mitogen for mesenchyme- and glia-derived cells. It consists of two disulfide-bonded polypeptide chains, A and B, and occurs as three isoforms, PDGF AA, PDGF AB and PDGF BB. The three isoforms bind with different affinities to two receptor types, A and B, which are structurally related and endowed with protein-tyrosine kinase domains. Ligand binding induces activation of the receptor kinases by formation of receptor dimers, the A subunit of PDGF binds only to A receptors with high affinity, whereas the B subunit can bind to both A and B receptors. Evidence suggests that PDGF may function as a neurotrophic factor. The fact that PDGF-A receptors are expressed in oligodendrocyte progenitor cells, whereas PDGF-B receptors are expressed on neurons, suggests that the different isoforms of PDGF may regulate growth and differentiation of different cell types in the developing central nervous system by paracrine and autocrine routes.Synonyms: PDGF A, PDGF alpha, PDGF subunit A, PDGF-1, PDGF-A, PDGF1, Platelet-derived growth factor A chain, Platelet-derived growth factor alpha, Platelet-derived growth factor subunit A
