  • Application Notes
    Approved: ELISA (0.5 - 1 μg/mL), Flo (50 μg/mL), WB (1 - 2 μg/mL)

    Usage: Suitable for use in Flow Cytometry, Western Blot and Direct ELISA. Flow Cytometry: 50 μg/mL, For intracellular staining to detect human Ret, cells must first be fixed and permeabilized using 4 % paraformaldehyde and 0.1 % saponin. Dilute this antibody to 50 μg/mL and add 10 μL of the diluted solution to 1-2.5x10^5 cells in a total reaction volume not exceeding 200 μL. The binding of unlabeled monoclonal antibodies may be visualized by adding 10 μL of a 25 μg/mL stock solution of a secondary developing reagent such as goat anti-mouse IgG conjugated to a fluorochrome. Cells should be washed for a final time in 0.1 % saponin prior to flow cytometric analysis. Western Blot: 1-2 μg/mL, Detection limit for rhRet is ~50 ng/lane under non-reducing and reducing conditions. Use chemiluminescence to increase sensitivity. Direct ELISA: 0.5-1.0 μg/mL, The detection limit for rhRet and rmRet is ~6 ng/well.
