Cdt1 and its inhibitor Geminin are important regulators of replication licensing. A critical balance between both proteins functions to ensure that each origin only fires once during the cell cycle. Cdt1 cooperates with Cdc6 to promote the loading of the MCM complex onto chromatin to form the pre-replication complex. Overexpression of Cdt1 leads to aberrant replication and activation of DNA damage checkpoints.
