  • Application Notes
    Flow Cytometry: 0.5-1 μg/million cells in 0.1ml

    Immunofluorescence: 0.5-1 μg/mL

    Western Blotting (0.25-0.5 μg/mL)

    Immunohistochemistry (FFPE): 0.5-1 μg/mL for 30 minutes at RT (1)

    Prediluted format : incubate for 30 min at RT (2)

    The optimal dilution of the HSP27 antibody for each application should be determined by the researcher.

    1. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10  mM Citrate Buffer,  pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.
    2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

    Background
    It recognizes a 24-27  kDa estrogen-regulated protein, identified as heat shock protein 27 (hsp27). Hsp27 was recently found to be identical to the estrogen-induced 'p29' and '24K' protein. About 50 % of breast carcinomas are positive for hsp27 especially those that are also positive for estrogen and/or progesterone receptor. HSP27 has also been implicated in drug resistance in cancer cells.
