Specificity: This antibody reacts with a 110 kD human cell surface glycoprotein known as HNK-1. This CD marker recognizes peripheral blood lymphocyte subsets and natural killer (NK) cells. Many cells which co-express CD57 and CD8 antigens are a subset of suppressor/cytotoxic T-cells. These antibodies recognize neuroendocrine cells and their tumors. CD57 antigens are not expressed on erythrocytes or platelets.

Purification: Concentrated.

Immunogen: Human peripheral blood mononuclear cells.

Application Notes: Flow cytometry: (not tested at our lab.)
Immunohistochemistry: 1:50-1:100.
Staining Procedure: This antibody can be used on frozen tissue section and on formalin-fixed, paraffin-embedded tissue sections. Prolonged fixation in buffered formalin can destroy the epitope. The antibody may be used at a dilution of 1:501:100 in IHC with predigestion or high temperature antigen unmasking technique (Boiling the tissue in 10 mM citrate buffer for 10-20 mins. followed by cooling at RT for 10-20 mins) may enhance staining on formalin fixed tissues. The optimal conditions should be determined by the individual laboratory.
