Immunohistochemistry on Paraffin Sections: 1/25-1/75 in an ABC method (30 minutes atroom temperature)Recommended Positive Control: Tonsil. Formalin fixed paraffin embedded tissue sections require high temperature antigenunmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

IL-2R alpha is over expressed on the surface of activated T cell, HTLV-1 or HTLV-2 positive T cell lines and moderately expressed on activated B cells, NK-cells and macrophages. The Interleukin 2 Receptor alpha and beta chains, together with the common gamma chain, constitute the high affinity IL2 receptor present on activated T and B cells, thymocyte subset, pre B cells and T regulatory cells. Homodimeric alpha chains result in low affinity receptor, while homodimeric beta chains produce a medium affinity receptor. Normally an integral membrane protein, soluble IL2 Receptor alpha has been isolated and determined to result from extracellular proteolysis.
