Application Notes: Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhLeptin (7.5 ng/mL), a concentration of 0.5 - 3.0 μg/mL of this antibody is required. ELISA: To detect hLeptin by direct ELISA (using 100l/well antibody solution) aconcentration of at least 0.5g/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 -0.4 ng/well of recombinant hLeptin. Western Blot: To detect hLeptin by Western Blot analysis this antibody can be used at aconcentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hLeptin is 1.5 - 3.0 ng/lane, under either reducing ornon-reducing conditions. Immunohistochemistry: use at approx. 2 μg/mL for cryostat and paraffin sections.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.

Background: Leptin plays a critical role in the regulation of body weight by inhibiting food intake and stimulating energy expenditure. Defects in Leptin production cause severe hereditary obesity in rodents and humans.
