Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhIL-12 (0.3 ng/mL), a concentration of 0.012 - 0.02 μg/mL of this antibody is required. ELISA: To detect hIL-12 by direct ELISA (using 100 l/well antibody solution) aconcentration of at least 0.5g/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with compatible secondary reagents, allows the detectionof 0.2 - 0.4 ng/well of recombinant hIL-12. Western Blot: To detect hIL-12 by Western Blot analysis this antibody can be used at aconcentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hIL-12 is 1.5 - 3.0 ng/lane, under either reducing ornon-reducing conditions. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Store the antibody prior to reconstitution at -20 °C. Following reconstitution the antibody can be stored at 2-8 °C for one month or at -20 °C for longer.
Interleukin 12 (IL12) can act as a growth factor for activated T and NK cells, enhance the lytic activity of NK/lymphokine activated killer cells, and stimulate the production of IFN gamma by resting PBMC. IL12 induces IFN gamma, TNF alpha production in T and NK cells, costimulation of PBL proliferation, proliferation/differentiation of TH1 T lymphocytes. It is downregulated by IL10, homodimeric p40 antagonistic to functional p70 heterodimer.
