The antibody has been tested by ELISA and Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:500 ~ 1:2,000. This antibody (5C2) has the specificity against the NAC domain (61-95aa) of alpha-synuclein. Recommended starting dilution is 1:1,000.

Alpha-synuclein (amino acids 1-140), an acidic neuronal protein of 140 amino acids, is extremely heat-resistant and is natively unfolded with an extended structure primarily composed of random coils. a-synuclein has been suggested to be implicated in the pathogenesis of Parkinson's disease and related neurodegenerative disorders, and more recently, to be an important regulatory component of vesicular transport in neuronal cells. Specially, the non-Abeta component of amyloid plaques (NAC, amino acids 61-95) is a highly amyloidogenic peptide consisting of 35 amino acids which was first identified associated with senile plaques in the Alzheimer's disease brain. Clone: Anti-human alpha-Synuclein mAb, clone 5C2, is derived from hybridization of mouse SP2/0 myeloma cells with spleen cells from BALB/c mice immunized with a recombinant human alpha-synuclein protein. This antibody specifically recognizes alpha-synuclein only, particularly the NAC domain of alpha-synuclein.
