Characteristics: The ICRF44 monoclonal antibody reacts with CD11b, the 165- kDa adhesion glycoprotein that associates with the 95- kDa integrin β2 (CD18) to form the CD11b/CD18 complex, also known as Mac-1 or CR3. CD11b is expressed on activated lymphocytes, monocytes, granulocytes, and a subset of NK cells. CD11b functions in cell-cell and cell-substrate interactions and is a receptor for iC3b, CD54 (ICAM-1), CD102 (ICAM-2) and CD50 (ICAM-3). This antibody significantly inhibits polymorphonuclear leukocyte aggregation in response to fMLP.

Purification: The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography.

Immunogen: Human monocytes.

Application Details

Application Notes: Immunohistochemistry: The ICRF44 (44) antibody is recommended to test for immunohistochemical staining of acetone-fixed frozen sections. Tissue tested was human spleen and tonsil. The antibody stains activated lymphocytes, monocytes, granulocytes and a subset of NK cells. The isotype control recommended for use with this antibody is purified mouse IgG1. For optimal indirect immunohistochemical staining, the ICRF44 (44) antibody should be titrated (1:10 to 1:50 dilution) and visualized via a three-step staining procedure in combination with polyclonal, biotin conjugated anti-mouse Igs (multiple adsorbed) as the secondary antibody and Streptavidin-HRP together with the DAB detection system. The clone ICRF44 (44) is not recommended for formalin-fixed paraffin embedded sections.
