- Characteristics
- This antibody recognizes a single protein of 33-34 kDa, identified as the Prostate specific antigen. This PSA antibody is highly specific and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. Prostate specific antigen is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign hyperplasia.

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the PSA antibody to be titered up or down for optimal performance.
1. Staining of formalin-fixed tissues requires boiling tissue sections in 1 mM EDTA Buffer, pH 7.5-8.5, for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.\. FACS: 0.5-1 μg/million cells,IF: 0.5-1 μg/mL,IHC (FFPE): 0.5-1.0 μg/mL for 30 minutes at RT (1),Prediluted format : incubate for 30 min at RT (2) - Restrictions
