  1. Data suggest that hepatic glucokinase (show GCK Antibodies) activity is regulated by reversible binding to specific inhibitor protein glucokinase regulatory protein (GKRP) and by binding to activator proteins such as 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase (PFK2 (show PFKFB3 Antibodies)/FBP2 (show KHSRP Antibodies)); changes in glucokinase (show GCK Antibodies) expression and activity are associated with poorly controlled type 2 diabetes and nonalcoholic fatty liver disease. 2. The rs780094 [1.34 (1.21-1.49); p = 8.57 x 10(-8) ] on chr 2 at the glucokinase regulatory protein (GCKR) locus was similarly significantly associated, while the rs10911205 [1.29 (1.16-1.44); p = 3.52 x 10(-6) ] on chr1 at the laminin subunit gamma-1 (LAMC1 (show LAMC1 Antibodies)) locus showed suggestive association with disease.

GCKR Antigen Profile

Antigen Summary

This gene encodes a protein belonging to the GCKR subfamily of the SIS (Sugar ISomerase) family of proteins. The gene product is a regulatory protein that inhibits glucokinase in liver and pancreatic islet cells by binding non-covalently to form an inactive complex with the enzyme. This gene is considered a susceptibility gene candidate for a form of maturity-onset diabetes of the young (MODY).
