ELISA: (direct or indirect: 1:500-1:5,000). Flow Cytometry: (1:20-1:100). Immunohistochemistry: (paraffin sections). Western blot: (1:500-1:5,000 using ECL. Suggested blocking and dilution buffer is PBST with 0.05 % Tween 20 and 5 % skim milk. Suggested incubation time is 1 hour at rooom temperature). Jagged-1 is a ligand for multiple Notch receptors and involved in the mediation of Notch signaling. It is involved in cell-fate decisions during hematopoiesis. Is involved in early and late stages of mammalian cardiovascular development. Inhibits myoblast differentiation. Enhances fibroblast growth factor-induced angiogenesis (in vitro). Defects in Jagged-1 are the cause of Alagille syndrome type 1 (ALGS1) and tetralogy of Fallot (TOF).
