Target Details PHLPP1 Application Details Handling Images
Specificity PHLPP antibody has been reported to cross reacts with beta-catenin, which may display 90kDa band. We would not recommend using this specific antibody for assays where signals may not be notable between PHLPP1 and Beta-Catenin.
Cross-Reactivity (Details) Based on sequence homology, this antibody may react with Orangutan, Rhesus Monkey, Gorilla, Chimpanzee, Tasmanian devil, White-tufted-ear marmoset, Crab-eating macaque, Naked mole rat, Gray short-tailed opossum,Northern white-cheeked gibbon.
Purification affinity purified
Immunogen The immunogen recognized by this antibody maps to a region between residue 1150 and the C-terminus (residue 1205) of human PH Domain and Leucine Rich Repeat Protein Phosphatase using the numbering given in entry NP_919431.1 (GeneID 23239).

Antigen
Alternative Name PHLPP (PHLPP1 Antibody Abstract)
Background Located on chromosome 18 and called PH domain Leucine-rich repeat ProteinPhosphatase (PHLPP, pronounced flip), the tumor suppressor is describedin the April 1, 2005 issue of the journal Molecular Cell. The scientists demonstrated thatPHLPP deletes a phosphate molecule, causing termination of cell-growth signaling by aprotein called Akt that controls the balance between cell growth leading to cancer andcell death that prevents tumor formation. PHLPP levels are markedly reduced in severalcolon cancer and gliobastoma human cell lines that had elevated Akt phosphorylation. Alternate Names: anti-PH Domain and Leucine Rich Repeat Protein Phosphatase antibody, anti-SCOPantibody, anti-Suprachiasmatic Nucleus Circadian Oscillatory Protein antibody,anti-PLEKHE1 antibody, anti-Pleckstrin Homolog Domain Containing antibody,anti-Family E (with Leucine Rich Repeats) Member 1 antibody.
Gene Symbol: PHLPP1
Gene ID 23239, 98432
NCBI Accession NP_919431
Pathways PI3K-Akt Signaling, Fc-epsilon Receptor Signaling Pathway, Neurotrophin Signaling Pathway

Application Notes Western Blot: Beta catenin band in Western blot is relatively less intense than PHLPP band. We do not recommend this antibody in Immunohistochemistry, Immunocytochemistry and Immunofluorescence due to its cross reactivity to Beta Catenin.
Recommended dilutions: ICC/IF (Negative), Immunoprecipitation 1-4 µg/mg of lysate, Western Blot 1:2000-1:10000, Immunohistochemistry (Negative)
Restrictions For Research Use only

Format Liquid
Concentration 0.2 mg/mL
Buffer Tris-buffered Saline and 0.1 % BSA, Sodium Azide
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
