Suitable for Immunohistochemistry (frozen sections: 4-8 μg/mL, 1/50-1: 100) and FACS. Suggested positive control: Human tonsil. Does not react on routinely processed paraffin sections.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user. - Protocol
- Protocol with frozen, ice-cold acetone-fixed sections: The whole procedure is performed at room temperature1. Wash in PBS2. Block endogenous peroxidase3. Wash in PBS4. Block with 10% normal goat serum in PBS for 30min. in a humid chamber5. Incubate with primary antibody (dilution see datasheet) for 1h in a humid chamber6. Wash in PBS7. Incubate with secondary antibody (peroxidase-conjugated goat anti mouse IgG+IgM(H+L) minimal-cross reaction to human) for 1h in a humid chamber8. Wash in PBS9. Incubate with AEC substrate (3-amino-9-ethylcarbazol) for 12min. 10. Wash in PBS11. Counterstain with Mayer's hemalum

- Background
- E-Cadherin, also called L-CAM (Liver Cell Adhesion Molecule) is an epithelial cell adhesion molecule and plays an important role in the formation of cell-cell contacts in epithelia irrespective of their origin as ecto-, meso- or endodermal tissue. This early adhesion event between epithelial cells is a prerequisite for the assembly of all elements of the junctional complex and is as such important in maintaining epithelial integrity. Over 90 % of malignancies are carcinomas. One of the prerequisites for the release of carcinoma cells from the primary site might be a defect in intercellular adhesion mediated by the absence of E-cadherin expression. Therefore, the expression of E-cadherin might be an important parameter for the determination of the invasive potential of epithelial neoplasms, and for the transition of a benign to a malignant neoplasm.Synonyms: CAM 120/80, CDH1, CDHE, E-cadherin, Epithelial cadherin, UVO, Uvomorulin
