    Phospho-specific STAT3 selective antibodies were generated against a peptide taken from the human protein. The PSTAT3-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The phospho-specific STAT3-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The supplier provides PSTAT3 Western blot positive control samples in ready-to-use SDS-PAGE sample buffer.     Background
    Signal transducers and activators of transcription (STATs) are a family of 7 transcription factors that form part of the (Janus Kinase) JAK-STAT signaling cascade. This cascade is the basis of the signal transduction mechanism for many cytokine receptors. STATs translocate to the nucleus once activated by phosphorylation by JAKs. Many factors regulate the JAK-STAT pathway including STAT dephosphorylation by phosphatases, altered nuclear import-export dynamics of STAT, and STAT gene activation antagonists such as SOCS (suppressors of cytokine signaling) and PIAS (Prot. Inhibitors of Activated STATs, 1). The STAT3 gene is part of a family of the STAT proteins that carry out a dual function: signal transduction and activation of transcription. STAT3 is widely expressed and becomes activated through phosphorylation on tyrosine as a DNA binding protein in response to a variety of stimuli such as EGF, IL-6, PDGF, IL- 2 and G-CSF. STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein plays a key role in many cellular processes such as cell growth and apoptosis. Abnormal STAT3 protein activation is identified in many cancers of the breast, prostate, and pancreas, as well as cancers of blood-forming cells (leukemia and lymphoma,. Phosphospecific antibodies are affinity-purified rabbit polyclonal or monoclonal antibodies, monospecific for a target protein that is phosphorylated on a specific tyrosine, threonine, or serine residue. STAT3 is activated by phosphorylation on Ser-727, which leads to dimer formation, nuclear translocation, and regulation of gene expression. Transcriptional activation is regulated by phosphorylation at Ser727 through the MAPK or mTOR pathways. Mutations in STAT3 cause of hyperimmunoglobulin E recurrent infection syndrome autosomal dominant (AD-HIES) also known as hyper-IgE syndrome or Job syndrome. The human Phospho-specific STAT3 gene maps to chromosome 17q21.31 and encodes a 512 amino acid protein with phosphorylation occurring at Serine 727.
